Abstract
|
|
|
Since vitamin A and its congeners (retinoids) have been implicated in the control of cellular proliferation and differentiation of tumor cells, the ability of retinoic acid (RA) to inhibit the growth and to induce the morphologic differentiation
of
a
human neuroblastoma cell line. LA-N-5, was investigated to develop an in vitro model to study the relationship between cell differentiation and tumorigenicity.
The growth inhibition was observed at 10E-8M of RA and showed to be concentration-dependent. RA also inhibited the ability of LA-N-5 cells to form anchorage0independent colonies in soft agar which is closely correlated with cellular
tumorigenicity.
Treatment of cells with RA or 12-O-tetradecanoyl phorbol-13-acetate (TPA) resulted in an increase in an increase in the expression of antigen against Mab390 and an decrease in the expression of antigen against Mab459. RA decreased N-myc
expression
in
cultured neuroblastoma cells.
These results suggest that the cellular differentiation of neuroblastoma cells induced by RA is closely related with the loss of tunorigenicity and decreased N-myc expression and LA-N-5 could be used as an in vitro model to study differentation
and
tumorigenicity of cancer.
